Free-ling amoebae (FLA) including <i>Acanthamoeba</i> spp., <i>Naegleria fowleri</i>, <i>Balamuthia mandrillaris</i> and <i>Sappinia pedata</i>, can cause opportunistic infections leading to severe brain pathologies. Human infections with pathogenic FLA have been increasingly documented in many countries. In Switzerland, thus far, the occurrence and distribution of potentially pathogenic FLA has not been investigated. Swiss water biotopes, including swimming pools, lakes, rivers and ponds, have now been screened for the presence of FLA, and assessment of their pathogenicity potential for a mammalian host has been undertaken. Thus, a total of 17 isolates were recovered by in vitro cultivation from these different aquatic sources. Characterization by sequence analysis of <i>Acanthamoeba</i> spp.-specific and 'FLA-specific PCR products amplified from 18s rDNA based on morphological traits, thermotolerance, and cytotoxicity towards murine fibroblasts yielded the following findings: <i>Echinamoeba</i> cf. <i>exundans</i> (3 isolates), <i>Hartmannella</i> spp. (3), <i>Vannella</i> spp. (4), <i>Protacanthamoebica</i> cf. <i>bohemica</i> (1), <i>Acanthamoeba</i> cf. <i>castellanii</i> (1) and <i>Naegleria</i> spp. (5). <i>B. mandrillaris</i> and <i>N. fowleri</i> did not range amongst these isolates. None of the isolates exhibited pronounced cytotoxicity and all failed to grow at 42 °C; therefore, they do not present any potential for CNS pathogenicity for humans.

This study assessed the role of picocyanobacterial photosynthesis in the induction of calcite precipitation. It aimed at establishing whether photosynthetic uptake of bicarbonate by <em>Synechoccoccus</em> cells leads to calcite nucleation. The precipitation of calcite was initiated by addition of previously washed cyanobacterial cells of <em>Synechococcus</em> strain PCC 7942 to solutions of calcium carbonate at different saturation levels with respect to calcite. Precipitation experiments were performed under controlled laboratory conditions in two set-ups: one in which photosynthesis was inhibited using a herbicide called Diuron and the other one in which photosynthesis was taking place. During the experiments, a pH meter monitored the pH and ion selective electrodes monitored concentrations of carbonate and calcium ions. The morphology of the precipitated crystals was analysed using Scanning Electron Microscopy. When the kinetics of calcium carbonate nucleation by the <em>Synechococcus</em> cells were compared for the two sets of experiments, there were very little differences. In fact, the induction times for precipitation reactions with photosynthesis were shorter due to the uptake of carbon dioxide. It is therefore, concluded that photosynthesis does not directly influence the nucleation of calcite at the surface of <em>Synechococcus</em> cells with sufficient supply of carbon dioxide, i.e. cells took up carbon dioxide and not bicarbonate. The microscopic observations, however, provided some evidence that picocyanobacterial cell walls act as a template for calcite nucleation.

Batch adsorption and dissolution experiments with lepidocrocite (γ-FeOOH) and two siderophores, desferrioxamine B (DFOB) and aerobactin, were performed between pH 3 and 8 in the dark and under irradiation with UV-visible light. The increase in surface concentrations of adsorbed DFOB with increasing pH was explained in terms of electrostatic interactions between the protonated and charged terminal amine group of DFOB surface complexes and the charged lepidocrocite surface. The adsorption of aerobactin was consistent with the typical anion-like adsorption behavior of low molecular weight organic acids and indicated that the adsorption properties are strongly determined by the carboxylic acid groups. The adsorption experiments revealed furthermore that the Fe(III)-DFOB solution complex has a very low affinity for the surface, in contrast to Fe(III)-aerobactin solution complexes. In accordance with a surface-controlled mechanism of ligand-promoted dissolution, we found a linear correlation between dissolution rates of lepidocrocite and the surface concentrations of adsorbed DFOB. In the dark, 6- to 8-fold lower dissolution rate coefficients were determined for aerobactin in comparison to DFOB. These results suggested that aerobactin forms Surface complexes that are less dissolution-active, characterized by a higher degree of multinuclear surface complexation and/or by less dissolution-active coordination modes of the involved iron-binding groups. For both DFOB and aerobactin, dissolution rate coefficients increased significantly under irradiation with UV–visible light. This increase was interpreted in terms of light-induced reduction of surface Fe(III), primarily by intrinsic photochemical processes of the lepidocrocite bulk phase, based on the observed photoreductive dissolution in the absence of organic ligands between pH 3 and 6. We hypothesize that the α-hydroxycarboxylate group of aerobactin may form a surface complex that additionally promotes photoreductive dissolution by a ligand-to-metal charge-transfer (LMCT) reaction, similar to citrate. However, LMCT reactions involving the α-hydroxycarboxylate group of aerobactin are rather ineffective, based on the comparison of light-induced dissolution rate coefficients determined in the presence of aerobactin and citrate.

The adsorption of two model siderophores, desferrioxamine B (DFOB) and aerobactin, to lepidocrocite (γ-FeOOH) was investigated by attenuated total reflection infrared spectroscopy (ATR-FTIR). The adsorption of DFOB was investigated between pH 4.0 and 10.6. The spectra of adsorbed DFOB indicated that two to three hydroxamic acid groups of adsorbed DFOB were deprotonated in the pH range 4.0–8.2. Deprotonation of hydroxamic acid groups of adsorbed DFOB at pH values well below the first acid dissociation constant of solution DFOB species (p<i>K</i>_a = 8.3) and well below the point of zero charge of lepidocrocite (pH_PZC = 7.4) suggested that the surface speciation at file lower end of this pH range (pH 4) is dominated by a surface DFOB species with inner-sphere coordination of two to three hydroxamic acids groups to the surface. Maximum adsorption of DFOB Occurred at approximately pH 8.6, close to the first p<i>K</i>_a value of the hydroxamic acid groups, and decreased at lower and higher pH values.<br/> The spectra of adsorbed aerobactin in the pH range 3–9 indicated at least three different surface species. Due to the small spectral contributions of the hydroxamic acid groups of aerobactin, file interactions of these functional groups with the surface could not be resolved. At high pH, the spectral similarity of adsorbed aerobactin with free aerobactin deprotonated at the carboxylic acid groups indicated outer-sphere complexation of the carboxylate groups. With decreasing pH, a significant peak shift of the asymmetric carboxylate stretch vibration was observed. This finding suggested that the (lateral) carboxylic acid groups are coordinated to the surface either as inner-sphere complexes or as outer-sphere complexes that are strongly stabilized at the surface by hydrogen bonding at low pH.

Diesel and wood combustion are major sources of carbonaceous particles in the atmosphere. It is very hard to distinguish between the two sources by looking at soot particle morphology, but clear differences in the chemical structure of single particles are revealed by C(1s) NEXAFS (near edge X-ray absorption fine structure) microspectroscopy. Soot from diesel combustion has a dominant spectral signature at ~285 eV from aromatic <i>π</i>-bonds, whereas soot from wood combustion has the strongest signature at ~287 eV from phenolic carbon bonds. To investigate if it is possible to use these signatures for source apportionment purposes, we collected atmospheric samples with either diesel or wood combustion as a dominant particle source. No spectra obtained from the atmospheric particles completely matched the emission spectra. Especially particles from the wood dominated location underwent large modifications; the phenolic spectral signature at ~287 eV is greatly suppressed and surpassed by the peak attributed to the aromatic carbon groups at ~285 eV. Comparison with spectra from diesel soot samples experimentally aged with ozone show that very fast modification of the carbon structure of soot particles occurs as soon as they enter the atmosphere. Source attribution of single soot particles with microspectroscopy is thus hardly possible, but NEXAFS remains a powerful tool to study aging effects.

Fisheries research has discovered that the habitat situation in Swiss running waters, a parasitic fish disease and fisheries management are most likely responsible for declining fish catches in Switzerland. Regarding fisheries management, recreational fishermen (hereafter called anglers) can be considered as main stakeholders in Switzerland, who actively participate in management activities. Fish stocking (or stocking) is among the most common and widespread management tools. It can be defined as the intentional release of large numbers of fish derived from hatcheries, rearing streams or ponds into target rivers. Main motives for stocking are mitigation for human-caused habitat perturbations, restoration and conservation of stocks and harvest enhancement. While stocking is very popular among Swiss anglers and there is indication that they might overestimate its power, it is critically discussed among ecologists and fisheries scientists because it has the potential to threaten fish conservation and the sustainability of indigenous fish stocks through increased competition, loss of genetic distinctiveness and the spread of diseases and/or parasites. [...]<br /><br />In der Fischereiforschung wurde entdeckt, dass die Habitatsituation in Schweizer Fliessgewässern, eine parasitäre Fischerkrankung und Fischereimanagement höchst wahrscheinlich verantwortlich für abnehmende Fischfänge sind. Bezogen auf das Fischereimanagement können Hobby-Fischer (oder Angler) in der Schweiz als Hauptakteure bezeichnet werden, da sie aktiv in die Bewirtschaftung der Gewässer eingebunden sind. Eine sehr verbreitete und übliche Bewirtschaftungsmassnahme ist der sogenannte Fischbesatz (oder kurz: Besatz). Darunter wird das intentionale, meist im grossen Stil durchgeführte Aussetzen von Fischen in Zielgewässer verstanden. Die Besatzfische werden meist in Aufzuchtsbächen oder in Zuchtanlagen aufgezogen. Hauptmotive für Besatz sind die Verringerung von durch den Menschen verursachte Störungen des Lebensraums, Wiederherstellung von Fischstämmen, Arterhaltung angestammter Arten und die Erhöhung des eigenen Angelertrages. Besatzmassnahmen sind bei Schweizer Anglern sehr populär und es gibt Hinweise darauf, dass Angler den Nutzen von Besatz überbewerten. Andererseits betrachten Ökologen und anderen Wissenschaftler aus dem Bereich der Fischerei Besatzmassnahmen kritisch, da Besatzmassnahmen das Potential zugeschrieben wird durch Konkurrenz, den Verlust von genetischer und adaptiver Vielfalt und die Verbreitung von Krankheiten und/oder Parasiten angestammte Arten und deren Erhaltung zu bedrohen bzw. zu gefährden. [...]

Lakes are valuable natural resources. However, they are increasingly threatened by anthropogenic activities. Transport and mixing processes, which govern the distribution of chemical and biological constituents, play a crucial role in sustaining lake ecology. Understanding these complex processes can help us protect these precious resources and use them wisely.<br /> In order to better quantify the flux pathway of constituents in Lake Alpnach, in this study, mixing processes in Lake Alpnach were simulated by a two-dimensional model (CE-QUAL-W2). The model exhibited very strong capabilities in simulating large time scale events such as the second vertical seiche mode, temperature and velocity changes; this set a consistent starting point for further water-quality-oriented simulations. Nevertheless the model displayed weaknesses in predicting velocity fluctuations and small time-scale waves (&lt; about 7 hours), which may be source of errors in other lakes or in short time simulations.<br /> Vertical diffusivity (Heat-Budget Method) was also calculated in this study, in order to better characterize mixing. It was proven that reliable vertical diffusivity can be calculated from CTD data alone, although the coupling CTD-Thermistor chains will provide more accurate results.

In Nordeuropa wurde ein energieintensives System aufgebaut, das einerseits Ammonium und Nitrat für die Landwirtschaft herstellt, anderseits die gleichen Moleküle in der Kläranlage eliminiert. Da in häuslichem Abwasser nahezu 80% des gesamten Stickstoffs aus menschlichem Urin stammen, zielen neuartige Sanitärkonzepte auf Urintrennung und -verwertung ab, um den N-Kreislauf zu schliessen. Jedoch sind mehrere Pilotprojekte zu dem Ergebnis gekommen, dass die Stickstoffkonzentrationen in gelagertem Urin im Vergleich zu frischem Urin stark abnehmen. Dies haben Beobachtungen an der Eawag in Dübendorf bestätigt, wo im Jahre 2005 15 Trenntoiletten und 2 Lagertanks installiert wurden (NoMix-Anlage).<br /> Die Untersuchungen dieser Diplomarbeit haben ergeben, dass die Ausgasung von NH3 während des Transports und der Lagerung für 30-60% Stickstoffverlust verantwortlichist. Der Kamineffekt treibt Ammoniak aus der natürlichen Belüftungsleitung heraus. Dieses Gas wurde mit Hilfe von „Impinger“ erfasst und sein Massenstrom in Beziehung zur Aussentemperatur gesetzt, um die Ausgasung über Zeit modellieren zu können. Als Beitrag zur Konzentrationsabnahme spielt auch die Verdünnung mit Spülwasser eine Rolle. Diese beruht auf einer Designschwäche der eingesetzten Trenntoilette, die bei Anwenderfehlbenutzung Spülwasser durch ein Ventil zum Urintank fließen lässt. Die Bilanz der Stickstofffracht zeigt die Schwäche der bestehenden NoMix-Anlage hinsichtlich der Urinwiederverwertung auf. Im Kapitel "Systembeurteilung" werden leichte  Umbaumöglichkeiten aufgezeigt, die die Effizienz dieses Abwassertrennkonzeptes steigern und den Eintrag von Ammoniak in der Umwelt mindern können.<br /><br /> In Northern Europe industrial food production relies on synthetic nitrogen fertilizer, the massive application of which has adverse effects on the environment. On the other hand the same compounds naturally present in urban wastewater are removed in expensive treatment plants. Urine source separation technology seems to be a promising concept that could recover 80% of nitrogen from the urban wastewater and close the nutriment cycle. However, a significant number of pilot projects showed that the nitrogen concentration of the stored urine is much lower than expected. The EAWAG confirmed the same finding through investigation of the installed urine collecting system, composed of 15 NoMix toilets and 2 collecting tanks.<br /> The analysis of this diploma thesis shows that the ammonia volatilisation during the transport and storage of the urine is largely responsible for the low concentration in the tank. Between 30 and 60 % of total nitrogen input gas out of the ventilation pipe,depending on the outdoor temperature (stack effect). The ammonia flow could be measured with “Impinger” and modelled over the time. The weak design of that toilet prototype combined with wrong use let flush water reach the tank and enhance the decrease of concentration. The nitrogen balance reveals the low N-recovery of urine in that NoMix plant. The concluding remarks of this thesis point at improvements regarding the efficiency of the system and the ammonia diffusion in the environment.

The degradation of chitin by heterotrophic bacteria is an important process in the biogeochemical cycling of carbon and nitrogen. Samples from three Swiss lakes with variable trophic status were amended with dierent types of chitin to enrich their bacterial community utilizing chitin. Bacterial abundance of enrichments over 21 days was analyzed by ow cytometry. Hydrolysis of substrate analogue 4-methylumbelliferyl diacetyl-chitobioside (MUF-diNAG) was measured and chitinase A (<em>chiA</em>) gene copies were enumerated by quantitative polymerase chain reaction (Q-PCR) to assess chitinolytic activity of samples. The bacterial diversity was analyzed by ngerprinting communities with denaturing gradient gel electorphoresis (DGGE). Results show that enrichment of chitinolytic community is possible when amending lake samples with chitin from crab shell, glucosamine or chitin extracted from natural zooplankton. The bacterial abundance increased in all samples, so did the number of <em>chiA</em> gene copies. Hydrolysis of substrate analogue could only be detected in samples enriched on crab shell chitin. Enrichment of bacteria on chitin substrates resulted in bacterial community shifts toward lower species diversity and higher intensity of single species. Similar community composition was found for enrichments on crab shell chitin and glucosamine, but with dierent response rates. Enriched communities from the three dierent lakes shared high similarity even though their initial community compositions varied. Our results suggest that by chitin amendment of lake samples chitinolytic bacteria can be isolated. Their community composition is most likely inuenced by the type of chitin.

The matrix from periphyton was chemically characterized in order to better understand its role in metal interactions and bioaccumulation of metals in periphyton communities. Periphyton was grown on glass slides using natural river water in flowthrough channels. Sixteen samples, ranging from 17‐40 days old, were taken during a four‐month period and the matrix was extracted and analyzed. Liquid Chromatography‐Organic Carbon Detection‐Organic Nitrogen Detection (LC‐OCD‐OND) revealed that the matrix consists of several fractions: biopolymers, building blocks of humic substances, LMW acids, and LMW neutral compounds. Variations in the amount of these compounds were observed in the periphyton matrix and appeared to be dependent on periphyton age. Bradford, phenol‐sulfuric acid, and amino acid assays were performed to quantify the amount of protein, carbohydrate, and amino acid in the matrix, respectively. Findings showed that the matrix is comprised of approximately half protein, half carbohydrate with relatively less free amino acid. ICP‐MS analyses indicated the presence of both essential and non‐essential trace metals in the matrix. Cd, Pb, Cr, Ni, Cu, Zn, Mn and Fe were measured, listed in increasing amount observed, yet were consistent during the observed 40 days of periphyton growth. Low values of metals normalized to organic carbon indicate the presence of excess functional groups for potential metal binding.

Werkstoffe aus Kunststoff, die zur Verteilung oder Speicherung von Trinkwasser verwendet werden sollen und direkt mit Trinkwasser in Kontakt kommen können, müssen vor dem Einsatz hinsichtlich zweier Punkte geprüft werden: (1) auf den Einfluss des Materials auf den Gehalt an organischen Kohlenwasserstoffen infolge der Migration und (2) auf die Vermehrung von Mikroorganismen auf der Materialoberfläche. Für beide Punkte existieren in Europa Prüfmethoden. In der Norm EN 12873-1 ist eine Migrationsprüfung vorgeschrieben, in der nach drei Migrationsperioden im Migrationswasser des Probenmaterials der Gehalt des gesamten organischen Kohlenstoffs (TOC = total organic carbon) gemessen wird. Zur Quantifizierung des Biofilmbildungspotentials auf den Materialoberflächen werden verschieden Prüfmethoden in den Ländern Europas durchgeführt – eine einheitliche Norm hierfür existiert auf europäischer Ebene nicht. Der TOC als Prüfparameter alleine hat eine relative geringe Aussagekraft und lässt keine Rückschlüsse auf toxikologische oder mikrobiologische Eigenschaften der Migrationssubstanzen zu. Die Methoden zur Abschätzung der Biofilmbildung sind alle sehr langwierig und können mehrere Monate dauern. An der Eawag wurde deshalb auf Grundlage der EN 12873-1 ein Migrationstest entwickelt, in dem durch die Messung der Konzentration des TOC und des assimilierbaren organischen Kohlenstoffs (AOC = assimilable organic carbon) im Migrationswasser chemische und mikrobiologische Messparameter gemessen und miteinander verglichen werden können. Durch Bestimmung des AOC, der eine durch Mikroorganismen leicht abbaubare Fraktion des TOC ist, wird es möglich, den TOC auch von der biologischen Seite genauer zu charakterisieren. In dieser Arbeit wurden Rohrleitungs- und Dichtungsmaterialien getestet, die vorwiegend im Hausinstallationssystem eingesetzt werden. Die Ergebnisse zeigten, dass sich die Materialien in ihrem Migrationsverhalten und in der mikrobiologischen Verwertbarkeit der Migrationssubstanzen deutlich unterschieden.<br /> Ein grosser Vorteil der neuen Testmethode gegenüber den Standardverfahren ist die kurze Durchführungszeit von maximal 3 Wochen. Mit Hilfe des entwickelten Migrationstests ist es nun möglich, eine schnelle und sichere Aussage über den Einfluss von Kunststoffmaterialien auf die chemische und mikrobiologische Qualität eines Trinkwassers treffen zu können.

Nowadays, mobile phone antennas can be found almost everywhere. We can see many of them in urban regions and also in very remote areas. Where the service provider is lacking a sufficient cable network, point-to-point radio systems are normally used to connect one antenna to the next or to the a nearby access point of the telephone network. The network of radio links has become very dense in the last years, especially in urban areas. But this kind of transmission is only possible if there is direct line of sight between the sender and the receiver. As soon as it starts to rain, the signal strength is lowered because the rain drops hamper the propagation of the electromagnetic waves. This attenuation of the signal depends on the intensity of the rainfall and can be measured at the receiver. By means of a widely accepted empirical relationship it is possible to estimate the average rain rate along the link from the attenuation measurements at the receiver.<br /> An urban area of about 150km² in Greater Zurich is subject to this thesis. Measurements from 23 microwave links are provided by a local network operator and reference data from 13 rain gauges, two disdrometers and a weather radar were available. A total of 128 calibrations were conducted to estimate the parameters for the empirical relationship and two different types of the necessary pre-processing of the data were compared to each other. It could be shown that 50 per cent of the predictions made by means of a constant parameter set, are afflicted with a mean error of less than 1.2mm/h. Differences in the drop size distribution of rain events and the quality of the received signal were identified as important influence factors for reliable model predictions.<br /><br /> Mobilfunkantennen findet man heutzutage fast überall. Wir begegnen vielen davon in den städtischen Gebieten aber auch in abgelegenen Regionen. Überall dort, wo es den Mobilfunkanbietern an einem ausreichend dichten Kabelnetz mangelt, wird Richtfunk eingesetzt um die einzelnen Antennen mit einander zu verbinden und ans Telefonnetz anzuschliessen. Dieses Netz von Richtstrahlverbindungen ist in den letzten Jahren vor allem in den städtischen Gebieten immer dichter geworden. Diese Art der Informationsübertragung ist jedoch nur möglich, wenn zwischen dem Sender und dem Empfänger eine direkte Sichtverbindung besteht. Sobald es zu regnen beginnt, wird das Signal abgeschwächt, da die Regentropfen die Ausbreitung der elektromagnetischen Wellen behindern. Diese Dämpfung des Signals hängt von der Niederschlagsintensität ab und kann am Empfänger gemessen werden. Mithilfe einer gängigen empirischen Beziehung ist es möglich, aus den Dämpfungsmessungen am Empfänger die mittlere Regenintensität entlang der Verbindung zu bestimmen.<br /> Diese Arbeit bezieht sich auf ein ungefähr 150km² grosses städtisches Gebiet im Grossraum Zürich. Es wurden Messungen von 23 Mikrowellenverbindungen eines lokalen Mobilfunkanbieters zur Verfügung gestellt und es waren Referenzdaten von 13 Regenschreibern, zwei Tropfenspektrometern und einem Wetterradar verfügbar. Insgesamt wurden 128 Kalibrierungen durchgeführt um geeignete Parameter für die empirische Beziehung zu schätzen und es wurden zwei unterschiedliche Varianten der nötigen Vorverarbeitung der Daten miteinander verglichen. Es konnte gezeigt werden, dass 50 Prozent der Vorhersagen, die mit einem konstanten Parameterset durchgeführt wurden, mit einem mittleren Fehler von weniger als 1.2mm/h behaftet sind. Unterschiede in den Tropfengrössenverteilung von Regenereignissen und die Qualität des gemessenen Signals wurden als wichtige Einflussfaktoren für zuverlässige Ergebnisse der Modellierung identifiziert.

In this Master's Thesis the use of dynamic time warping in the eld of wastewater treatment is presented.<br /> Dynamic time warping is a method to compare data signals. It is used to assign the influent data points to the euent data point of a reactor. The assignment is performed by constructing a cost-matrix and then nding the optimal way diagonally through this matrix.<br /> An optimal con guration of the dynamic time warping algorithm was found and implemented in Matlab.<br /> It could been shown, that with dynamic time warping an approximation of the mean of the hydraulic residence time distribution could be determined. With larger hydraulic residence times the absolute error increases. The dynamic time warping analysis allow to dermine the hydraulic residence time for non-steady flow conditions.<br /> With generated data it could been shown, that it is possible to obtain Nash-Sutcliffe coeffcients above 0.6 in representing the variability of the hydraulic residence time with dynamic time warping for ideal reactor models with a number of reactors higher two and mean hydraulic residence times smaller two hours.<br /> It could further been shown, that normalizing the data signals improves the dynamic time warping results for rst order reactions with reaction constants between 0 and -0:1 1/<em>hr</em>.<br /> With in measurement campaigns on the wastewater treatment plant Kloten/Opfikon collected and generated data, dynamic time warping was used to identify the hydraulic residence time in sewer and grit chambers successfully.<br /> For mean hydraulic residence times smaller one hour the distribution of the discharge on reactors operated in parallel was approximated that the assumption of equal hydraulic residence times is no longer necessairy.<br /> For the wastewater treatment plant Kloten/Opfikon a relation of the ratio of the residence times in the parallel grit chambers and the measured discharge could been derived.<br /> The results were validated with a tracer experiment in the parallel girt chambers and the primary clari ers. Through the left grit chamber and primary clari er an approximately 10 % higher discharge was passing while the tracer experiment was running by a discharge of 765:3 81:9 <em>m</em>³/<em>hr</em>.

Cichlids are one of the most species rich and diverse vertebrate families on the planet. However, like in many other species, this diversity is affected by anthropogenic influences. In Lake Victoria, it is hypothesized that the interaction between eutrophication and an invasive species (Nile Perch) has led to one of the largest mass extinctions ever recorded which mainly affected cichlid species living in the open water. Rock and sand dwelling species close to the shore were less affected. Die vorliegende Arbeit befasst sich mit der Thematik des Abbrechens eines durch gezielte Interventionen neu eingeführten Gesundheitsverhaltens namens SODIS in Bolivien. Solar Water Disinfection (SODIS) ist eine einfache Trinkwasser-Aufbereitungsmethode, welche auf Haushaltsniveau mit durchfallerzeugenden Bakterien verschmutztes Rohwasser trinkbar macht.<br /> Das Transtheoretische Modell und das Innovation-Decision-Process Model beschreiben den komplexen und stufenartigen Prozess einer Verhaltensänderung und dienen in der vorliegenden Arbeit als Erklärungsansätze. Die Konstrukte der Injunktiven und der Deskriptiven Norm werden ebenfalls untersucht, um das Abbrechen von SODIS erklären zu können.<br /> Das zentrale Forschungsziel dieser Arbeit liegt darin, verschiedene SODIS-Verhaltensuntertypen von Abbrechern und Anwendern zu identifizieren und bezüglich psychologischer und sozialer Variablen zu vergleichen. Für die eruierten Abbrecher- und Anwender-Typen sollen auf Grund der unterschiedlichen Ausprägungen in den relevanten Variablen Prognosen für einen wahrscheinlichen weiteren Verlauf innerhalb des Transtheoretischen Stufenmodells gestellt werden. Darüber hinaus werden Überlegungen zu möglichen Interventionen gemacht, welche einem Abbrechen zu einem früheren Zeitpunkt entgegenwirken oder einen möglicherweise bevorstehenden Abbruch verhindern könnten.<br /> Die Resultate der statistischen Auswertungen der mit Fragebogen erhobenen Daten belegen eindeutige Unterschiede zwischen den vier Verhaltenstypen, die mittels Cluster-Analyse identifiziert wurden. Es wurden gefunden: Der Abbrecher-Typ 1 scheint SODIS schon früh nach Beendigung der Interventionsphase nicht mehr anwenden zu wollen und bevorzugt es, sein Trinkwasser abzukochen. Der Abbrecher-Typ 2, der erst später mit SODIS aufhört, hat früher verhältnismässig viel Wasser mit Hilfe von SODIS aufbereitet und gedenkt auch, dies in Zukunft wieder zu tun. Erschwerend für diesen Verhaltenstyp scheint zu sein, dass er seine externen Erinnerungshilfen verloren hat. Der Anwender-Typ 1 ist SODIS gegenüber zwar positiv eingestellt, aber durch die vergleichsweise eher geringe Gewohnheit und Intensität mit der er SODIS bisher anwendete, besteht die Gefahr, dass er in naher Zukunft abbricht. Anwender-Typ 2 hingegen scheint das Verhalten SODIS tatsächlich angenommen zu haben. Es wird deshalb nicht erwartet, dass er es in nächster Zeit wieder aufgeben wird.

During the last century, human activities have added an impressing amount of nitrogen (N) to the environment and thereby severely changed the global N cycle (Saunders and Kalff 2001). Aquatic systems such as rivers and lakes are overloaded with N (mainly NO₃^-), which next to phosphorous leads to eutrophication and acidification of the system. Moreover, elevated NO₃^- concentrations can have a negative impact on human health. Rivers and streams represent an essential sink for bioavailable N as they are tightly linked to the terrestrial ecosystem and exhibit high biological uptake of nutrients.<br /> Denitrification is one of the most important processes permanently removing N from N-saturated rivers (Saunders and Kalff 2001; Seitzinger et al. 2006; Mulholland et al. 2008). Especially the hyporheic zone within floodplains exhibits high reaction rates and represents a high potential for C and N transformations (McClain et al. 2003). However, flow regulations of the past century such as constructions of levees and river straightening have reduced lateral connections among rivers, their floodplains and the hyporheic zone (Valett et al. 2005). River restorations may therefore be an important measure to increase such connections, and by this to simultaneously enhance the transformation potential for nutrients and organic matter. Microbial activity, such as denitrification, in groundwater and the hyporheic zone is often limited by bioavailable organic carbon (OC). Dissolved OC can be imported from regions of seasonal saturation into the hyporheic zone during an elevated water table (Valett et al. 2005; Forshay and Stanley 2005). Hence, the hydrological regime may be highly important for biogeochemical processes in the riparian zone. Unfortunately until this time, only little research has been conducted concerning N removal from the hyporheic zone. [...]<br /><br /> Im letzten Jahrhundert hat der Mensch eine beeindruckende Menge an Stickstoff (N) der Umwelt zugefügt und so den globalen Stickstoffkreislauf stark verändert (Saunders und Kalff, 2001). Aquatische Systeme wie Flüsse und Seen sind mit N (hauptsächlich NO₃^-) überladen, was neben Phosphor zur Eutrophierung und Versauerung der Systeme führt. Zudem können erhöhte NO₃^--Konzentrationen einen negativen Einfluss auf die menschliche Gesundheit haben. Flüsse stellen eine wichtige Senke für biologisch verfügbaren N dar, da sie eng mit dem terrestrischen Ökosystem verknüpft sind und eine grosse biologische Aufnahmekapazität von Nährstoffen aufweisen.<br /> Die Denitrifikation ist eine der wichtigsten Prozesse, die N permanent von N-überladenen Flüssen entfernen kann (Saunders und Kalff 2001; Seitzinger et al. 2006; Mulholland et al. 2008). Vor allem die hyporheische Zone innerhalb des Überschwemmungsgebiet verfügt über hohe Umsatzraten und stellt ein hohes Potential für C- und N-Transformationen dar (McClain et al., 2003). Flussregulierungen aus dem letzen Jahrhundert, wie der Bau von Dämmen und Flussbegradigungen, haben jedoch die lateralen Verbindungen zwischen den Flüssen, ihren Überschwemmungsgebieten und der hyporheischen Zone reduziert (Valett et al., 2005). Flussrenaturierungen sind deshalb möglicherweise eine wichtige Massnahme, um solche Verbindungen zu erhöhen und dadurch gleichzeitig das Transformationspotential für Nährstoffe und organisches Material zu erhöhen. Mikrobielle Aktivität, wie Denitrifikation, ist im Grundwasser und in der hyporheischen Zone oft durch biologisch verfügbaren organischen Kohlenstoff limitiert. Gelöster organischer Kohlenstoff kann während eines erhöhten Wasserspiegels von Zonen, die saisonal saturiert sind, in die hyporheische Zone verfrachtet werden (Valett et al. 2005; Forshay und Stanley 2005). Demzufolge ist das hydrologische Regime möglicherweise für biogeochemische Prozesse im Uferbereich von Flüssen wichtig. Leider ist bis jetzt wenig Forschung betreffend N-Abbau aus der hyporheischen Zone durchgeführt worden. [...]

Despite being a very popular concept, there is a lack of empirical knowledge regarding the successes of implementing Intergraded Water Resources Management (IWRM). This study reveals how Mozambique adopted the IWRM concept and main principles into its national water law, policies and institutions. The adoption of the IWRM concept occurred in a top-down direction, from the international level to the country level via external influence. Three main factors accounted for the adoption of the concept by Mozambique: a) developments of the IWRM approach at the global and regional levels, b) the influence of the international community through different organizations, and to a lesser extent c) the natural physical and socio-economic characteristics of the country. The IWRM principles have been initiated in the Limpopo River Basin, and despite all best efforts, the principles have been so far poorly implemented.

Decentralized systems which treat separated wastewater streams according to their specific properties are quite promising according to the newest research results. The use of a microbial electrolysis cell (MEC) to treat urine and produce hydrogen gas is such a novel technology. The MEC consists of two chambers which are connected via a cation exchange membrane. By applying voltage, the oxidation of organic substrate (assisted by exoelectrogens) at the anode can be coupled with a proton reduction at the cathode. EAWAG takes part in the investigations and set a MEC into operation in February 2009. The goal of this Master project is to understand and quantify the influence of some of the most important process parameters such as applied voltage, temperature and pH.<br /> The chronoamperometry measures the current at stepwise higher applied voltages. The influence of the applied voltage on the system performance was linear before reaching slowly a limit. The needed minimum voltage was determined to 238 mV. Those chronoamperometric curves gave indications that the system got diffusion limited with time. The coulombic efficiency represents the ratio between produced hydrogen according to the measured current and the theoretical possible hydrogen production calculated with a COD balance. The low coulombic efficiency of 57.7% leads as well to the assumption, that other bacteria degrade the organic substrate. Once the MEC was opened and a biofilm consisting of two layers could be observed: the wanted exoelectrogens and undesired other bacteria. By analyzing samples from the anode and the cathode chamber the sulphate degrading bacteria could be quantified to contribute 3% to the COD-consumption. Additional measures such as installing an oxygen-sensor to judge the possibility of aerobic growth or off-gas measurements to quantify the methanogenic growth should be considered. After classifying the bacteria, parameters to damage them uniquely should be investigated.<br /> The temperature experiment presented a linear increase in performance of 2.3% per degree. As the system is probably diffusion limited, the effect was caused by a bigger diffusive flux due to a temperature-dependent diffusion coefficient. The reaction at the cathode was not limiting at a pH between 6 and 9. When the pH was changed and the anode potential was fixed, the measured current increased 25% per pH unit. The growth curve of the exoelectrogens depends therefore linearly on the pH. If the applied voltage was constant, the current increase reached a limit, as due to the higher pH, the anode potential decreased. Sample analysis from the anode and cathode chamber proved, that the proton transport via the membrane is insignificant. The pH is therefore controlled in both chambers by adding sodium hydroxide solution or hydrochloric acid. If some day a perfect proton-exchange membrane is developed, proton-gradients might play a role and the before mentioned results from the pH-experiments might change. Nowadays, instead of adding chemicals, the effluent of the anode could be connected to the influent of the cathode. At the same time, this would unfortunately decrease the hydrogen purity. Some improvements are still needed, before the first full-scale reactor can be built. Another problem will be that resistances get bigger in a full-scale reactor. The measured resistances during the experiment decreased linearly with temperature and plotted against the applied voltage, a minimum resistance at 650 mV was found. For a full-scale reactor it would pay off to choose the operation parameters such that resistances get minimized.<br /> So there is still a long way to go before the first full-scale reactor will be set into operation. The biggest problems for the analyzed reactor at the moment are the growing competition and the insignificant proton-diffusion.

Nitrogen is the limiting nutrient in most wetlands and thus determines primary production and affects cosystem functions. Nitrogen cycling was studied in two tropical riverine wetlands in the Zambezi River Basin, Zambia, in October 08. To identify the dominant sources, sinks and internal processes in the nitrogen cycle nitrogen concentrations and speciation were determined. Nitrogen and carbon isotopic signatures were measured in various pools of the floodplain, namely dissolved and particulate organic matter in the main river channel, in soil, sediment, plants and periphyto. Acetylene reduction assays were conducted with soil, sediment, river water and periphyton samples to test for nitrogen fixation. Conceputal models and nitrogen budgets were developed and field and literature data was incorporated to futher narrow down dominatn processes. [...]

This research focused on implementation strategies of integrated river basin management (IRBM) and its related policy instruments. Policy instruments are wide-range tools used in order to steer or achieve specific objectives, in this case integration of the different sectors in water management within the natural boarders of a catchment area. This approach of IRBM is slowly infiltrating to authorities and current discussions are going on in Switzerland. The Netherlands on the other hand features broad and long experiences in (integrated) water management. Therefore it was assumed that the Netherlands applies more elaborated policy instruments towards integration than Switzerland. As a consequence, the main goal of this investigation was to get insights from the applied policy instruments in the Netherlands for the Swiss context that will support a future implementation of IRMB in Switzerland.<br /> For that reason the research approach of comparison study was chosen with a main focus on two catchment areas <em>Regge </em>in the Netherlands and <em>Birs </em>in Switzerland. In a first step, the applied policy instruments were detected and categorised in a theoretical conceptual frame. After this they were described by theoretical literature according to their characteristics, strengths and weaknesses. This outcome served further as basis for the examination of the policy instruments in practice, which was done by interviews of experts and literature review. In a last step the collected data was analysed having as base the empirical information; this was done with the objective of gaining insights and learned lessons from the Dutch practice for the Swiss context.<br /> It was found out that the different policy instruments feature rather different characteristics, which were therefore categorised in three classes according to their approach of achieving objectives: legislation and water management plans; financial instruments and contextbased instruments. Briefly it can be concluded that Switzerland can get several constructive insights from the Dutch policy instruments in IRBM related to legislation, water management plans, user charges, subsidies, information and implementation support, stakeholder participation, networking and benchmarking. No direct lessons can be transferred from the instruments of voluntary agreements.

Die Fliessgewässer der Schweiz haben in den vergangenen Jahrzehnten stark unter dem anthropogenen Druck gelitten. Eingriffe zwecks Hochwasserschutz, Landgewinnung und<br /> Stromerzeugung haben dazu geführt, dass viele Habitate zerstört wurden und die longitudinale Konnektivität entlang von Fliessgewässerkorridoren nicht mehr gewährleistet ist. Viele aquatische Organismen, allen voran die Fische, sind jedoch auf gesunde Habitate und durchgängige Fliessgewässer angewiesen. Heute wird vermehrt versucht, Migrationshindernisse wie Schwellen und Wehre zu entfernen und durch Rampen zu ersetzten, um den Fischen und anderen Organismen die Möglichkeit zurückzugeben, sich frei in den Gewässern zu bewegen. Ob solche Rampen jecloch wirklich funktionieren, ist oft nicht bestätigt worden. Eine Studie im Rahmen einer Dissertation soll mit Hilfe von Translokationsexperimenten klären, inwiefern solche Rampen für Fische durchgängig sind. Für Erfolgskontrollen von Rampen ist es sinnvoll, zu wissen, ob Fische nach Translokationen an die ursprünglich besetzten Stellen im Gewässer zurückkehren und welche Faktoren die Rückkehr beeinflussen. Zudem ist abzuklären, wie stark Fische in einem Gewässer an ihren Standort gebunden sind und nach einer Translokation (Hochwasser, Verfrachtung durch Menschen) wiederum exakt an diesen Ort zurückkehren.<br /> An zwei kleinen Fliessgewässern (Rykenbach und Ron) im Kanton Luzern wurde die Standorttreue und das Mikrohoming (die Rückkehr von Fischen an die vormals besetzte Position) an Bachforellen (Sa/mo trutta fario) und Alet (Leuciscus cepha/us) untersucht. Als inverses Mass für die Standorttreue wurde die lineare Grösse der Homeranges berechnet. Zur Ermittlung der Homerangegrösse wurden die Fische individuell markiert und mit Elektrobefischungen wiederholt gefangen. Bei den Bachforellen konnte festgestellt werden, dass mit zunehmender Fischlänge die Grösse der Homeranges signifikant abnahm. Bei den Alet jedoch nahm die Grösse der Homeranges mit zunehmender Fischlänge signifikant zu. Um das Mikrohoming zu untersuchen, wurden die Fische ebenfalls individuell markiert, über Distanzen von 150-400 m transloziert und mit Hilfe von Elektrobefischungen wiederholt gefangen. Dabei sollte festgestellt werden, welche der Faktoren (Fischlänge, Translokatonsrichtung, Translokationsdistanz und Jahreszeit) einen Einfluss auf das Mikrohoming ausüben. Nach der Translokation sind 27.1 % der Bachforellen und 38.2 % der Alet zurückgekehrt. Mit zunehmender Körperlänge der Bachforellen nahm auch die Wahrscheinlichkeit erfolgreichen Mikrohomings zu, sowie die Wahrscheinlichkeit einer schnelleren Rückkehr. Bei den Alet konnten keine Einflüsse der untersuchten Faktoren auf das Mikrohoming und die Geschwindigkeit der Rückkehr festgestellt werden. Auch konnten Unterschiede in der Habitatnutzung von juvenilen und adulten Bachforellen festgestellt werden. Adulte Bachforellen halten sich bevorzugt in tieferen Wasserzonen auf (z.B. Kolken), während sich juvenile Forellen in seichteren Wasserzonen und anderen hydraulischen Habitaten wie Gildes und Riffles aufhalten.<br /> Generell war von Interesse, wie sich Bachforellen und Alet nach einer Translokation verhalten und ob sie ein Mikrohoming zeigen. Beide Arten sind relativ stark ortsgebunden und ein grosser Anteil beider Arten kehrt an die ursprünglich besetzte Position zurück. Beide Arten sind sehr wohl in der Lage, nach einer Translokation zurückzukehren.<br /> Bei Translokationsexperimenten zwecks Funktionsüberprüfung von Rampen kann nun ungefähr abgeschätzt werden, in welchem Ausmass eine Rückkehr der Fische zu erwarten ist. Faktoren, die eine Rückkehr beeinflussen, könnten die Interpretation von Rampenexperimenten erschweren. Deshalb sollte zumindest der Einfluss der Körperlänge der zu untersuchenden Art ermittelt und bei der Interpretation der Resultate miteinbezogen werden.

Alpine waters are changing rapidly in response to climate change. Models predict not only general warming, but also an increase in extreme weather events. Since alpine aquatic insects are already living in relatively extreme environments, they may respond quickly to environmental changes. Extreme weather events such as droughts could lead to bottlenecks in alpine aquatic insects that must pupate and emerge before the drying of streams. Such extreme events and the resulting bottlenecks are likely to impact population genetic structure. Since alpine aquatic insects often have insular populations, this impact on population genetic structure may be even more pronounced than in closely related lowland species. [...]<br /><br /> Alpine Gewässer reagieren schnell auf Veränderungen des Klimas. Verschiedene Modelle sagen nicht nur eine allgemeine Erwärmung voraus, sondern auch eine Zunahme extremer Wetterereignisse. Da alpine aquatische Insekten sowieso schon in extremen Habitaten leben, könnten diese schnell auf Veränderungen in ihrer Umwelt reagieren. Extreme Wetterereignisse wie Dürren könnten zu Flaschenhälsen in aquatischen Insekten führen, die sich verpuppen und ausschlüpfen müssen bevor die Bäche und Flüsse austrocknen. Diese Extremereignisse und Flaschenhälse verändern wahrscheinlich die genetische Struktur der Population. Da alpine aquatische Insekten oft eine inselartige Struktur aufweisen, könnten die Auswirkungen auf die genetische Struktur dieser Populationen grösser sein als die Auswirkungen auf näher verwandte Arten in tieferen Lagen. [...]

Der dynamische Eintrag von polaren Pharmazeutika und Korrosionsschutzmitteln in aquatische Systeme erfolgt diffus oder aus Punktquellen. Ein hoher Probendurchsatz und ein über mehrere Einheiten variierender Messbereich sind Grundvoraussetzungen für die genaue Belastungsquantifizierung dieser Substanzen. Für deren Detektion in verschiedenen Umweltmatrizes ist die Hochleistungsflüssigkeitschromatographie-Tandem-Massenspektrometrie (HPLC-MS/MS) die Methode der Wahl. Um den routinemässigen Probendurchsatz zu erhöhen, ist ein automatisierter Anreicherungsschritt gekoppelt an die HPLC erstrebenswert. Das entscheidende Verbindungsglied dieses bestehenden Instrumentensystems ist die vollautomatische Anreicherung der Analyten.<br /> Die Herausforderung dieser Arbeit lag darin, diese in ihren chemischen und physikalischen Substanzeigenschaften so stark variierenden Substanzen mittels einer Methode im tiefen ng/L – Bereich detektieren zu können. Dabei gilt es, bezüglich der gegensätzlichen Substanzeigenschaften, ein Optimum für alle Substanzen betreffend der Anreicherungseffizienz auf dem geeigneten SPE (solid phase extraction) – Material zu erreichen. Dabei zeigte sich eine selbst entworfene Anreicherungskartusche für die Mehrheit der Substanzen, bestehend aus einem Mix an vier verschiedenen Materialien, am effizientesten. Weitere Systemparameter, wie die der Massenspektrometrie und Chromatographie, wurden im Zuge der Methodenentwicklung optimiert und nachfolgend integriert. Die Optimierung der chromatographischen Parameter erfolgte im Hinblick auf eine gute Ionisierungseffizienz der Analyten, sowie einer zufrieden stellenden Trennleistung.<br /> Die Validation der Methode ergab für mehr als 25 der zu quantifizierenden 30 Substanzen Nachweisgrenzen im Bereich von 0.5 – 10 ng/L. Damit erfolgt die Quantifizierung der Substanzen im tiefen ng/L – Bereich. Das selbst zusammengestellte Kartuschenmaterial zeigt für alle Substanzen sehr befriedigende Anreicherungseffizienzen von 70-130%, wobei die drei Umweltmatrizes darauf keinen nennenswerten Einfluss haben. Aufgrund der Validationsdaten findet die entwickelte Methode innerhalb eines Projektes zum Ab- und Umbau von Pharmazeutika innerhalb einzelner Aufbereitungsstufen einer Abwasserreinigungsanlage bereits Anwendung.<br /> Zusammenfassend lässt sich hervorheben, dass eine online SPE-HPLC-MS/MS-Methode zur Quantifizierung ausgewählter Pharmazeutika und Korrosionsschutzmitteln in Abwässern und Oberflächengewässern entwickelt wurde. Die Entwicklung eines starken analytischen Werkzeugs wurde realisiert, welches die zeitaufwendige offline SPE ersetzt und zudem eine hohe Sensitivität aufweist.

Seit etwa 15 Jahren werden Biozide in Fassadenbeschichtungen gegen Algen und Pilze eingesetzt. Einige dieser bioziden Wirkstoffe wurden in Oberflächengewässern nachgewiesen und es gibt Hinweise, dass relevante Mengen aus Siedlungsgebieten stammen - neben der Auswaschung beim landwirtschaftlichen Einsatz, wo teilweise dieselben Wirkstoffe verwendet werden. In dieser Arbeit wurde am Beispiel des Algizids Terbufryn untersucht, wie gross die auswaschbare Biozidmenge in Abhängigkeit von verschiedenen baulichen Strukturen ist. In Volketswil und Schwerzenbach wurden in den Industriezonen und drei VVohngebieten aus<br /> unterschiedlichen Bauzonen und -perioden die· biozidhaltigen Fassadenflächen bestimmt. Die entwickelte Methode zur Abschätzung der auswaschbaren Biozidmenge verband Felderhebungen und Berechnungen basierend auf GIS-Daten. [...]

In der Schweiz sind über 25 % der Fliessgewässer durch Hochwasserschutzbauten, hydro-elektrische Nutzung und Industrie stark beeinträchtigt, bzw. eingedolt. Dies entspricht über 15'800 km Gewässerlauf (BAFU 2006). Durch diese fortlaufenden Einwirkungen des Menschen auf die Fliessgewässer gehen immer mehr, für aquatische Lebensgemeinschaften wichtige Habitate verloren. lm Rahmen des Projektes „lntegra|es Flussgebietsmanagement“ (eine Zusammenarbeit von Eawag, Epfl, Wsl und Vaw) soll in der Schweiz abgeklärt werden, welchen Einfluss ausgewählte Habitatsparameter auf die Populationsstruktur und Biomasse von Bachforellen (Sa/mo trutta fario) sowie die Artenvielfalt der Fischfauna ausüben.<br /> Zu diesem Zweck wurden zwischen Mai und Oktober 2008 40 Fliessgewässerabschnitte in der Schweiz und im Fürstentum Lichtenstein im alpinen Bereich und im Mittelland beprobt. In einem ersten Schritt wurden die Gewässerabschnitte quantitativ befischt und anhand dieser Daten die Biomassen, Populationsstrukturen sowie die Artenzahlen der Fische ermittelt. In einem zweiten Schritt wurden dann zwölf aus der Literatur ausgewählte qualitative und quantitative Habitatsparameter erhoben (benetzte Breite, maximale Tiefe, Gefälle, Höhe über Meer, Fliessgeschwindigkeit, Beschattung, Verbauungsgrad Ufer, Kolmation, Substrat, Ufen/egetation, Mesohabitate Pool/Riffle/Glide/Cascade, Fischunterstandstypen).<br /> Die Habitatsparameter Gesamtfläche Fischunterstände, Shannonindex Fischunterstände, Beschattung, mittlere benetzte Breite und mittlere Fliessgeschwindigkeit hatten einen signifikanten Einfluss auf die Bachforellenbiomasse und -dichte. Die schrittweise multiple lineare Regression erzeugte eine Kombination von drei Habitatsparametern (Shannonindex Unterstände, Gesamtfläche Unterstände, Beschattung), die 84% der Varianz der Bachforellenbiomasse erklärten.<br /> Bei der Unterteilung der Bachforellendichte in die drei Altersklassen 0⁺, juvenil und adult erwiesen sich die Habitatsparameter Gefälle für die 0⁺-Bachforellendichte und mittlere maximale Tiefe für die adulten Bachforellendichte als signifikant. Es konnte hingegen kein Zusammenhang zwischen den ausgewählten Habitatsparametern und dem Variationskoeffizienten der Bachforellenlänge (als Indikator einer ausgewogenen Populationsstruktur) aufgezeigt werden.<br /> Es ergaben sich keine signifikanten Korrelationen zwischen der Fischdiversität und der Habitatsdiversität in Cyprinidengewässern_ Mit Hilfe einer „Canonical Correspondance Analysis“ konnte jedoch die positive Wirkung der Habitatsparameter Wasserpflanze (Unterstand) und Mesohabitat Glide auf die Dichte von in Cyprinidengewässern lebenden Arten nachgewiesen werden.<br /> Diese Masterarbeit zeigt, wie ausgeprägt sich bestimmte Habitatsparmeter (Gesamtfläche und Diversität der Fischunterstände, Beschattung, Tiefe, Breite, Fliessgeschwindigkeit, Gefälle) auf die Biomassen und Populationsstrukturen von Bachforellen auswirken können und weist auf die Wichtigkeit hin, diesen Habitaten bei Revitalisierungsmassnahmen besondere Beachtung zu schenken. Jedoch sollten zuerst in jedem Gewässer die relevanten Prozesse wiederhergestellt werden und die Wechselwirkungen zwischen Habitaten und Fischpopulationen genau untersucht werden, bevor konkrete Massnahmen getroffen werden. Dies ist wichtig, damit die künstlich hinzugefügten Habitate überhaupt wieder kolonisiert werden können (RONI ET Al.. 2002). Um den Zustand von Bachforellenhabiten schnell und zuverlässig beurteilen zu können, hat sich der Complexity index nach KERSHNER ET AL. (1997) als nützlich enıviesen.

The Zambezi River Basin (ZRB) faces future challenges in integrated water resourcemanagement (IWRM) due to rapid growth of population and economy. Tools areneeded to support IWRM decision making. A basin wide biogeochemistry modellays a solid foundation for further tool development. In this study, the Soil and Water Assessment Tool (SWAT) was applied to model basin wide biogeochemistry.The model (ZRB1) was calibrated and a local sensitivity analysis conducted. Ni-trogen and phosphorous exporting patterns in the ZRB were analyzed and criticallyevaluated. The prediction quality of ZRB1 in terms of flow and nutrient loadingswas found insufficient. By discussing possible shortcomings of the model, this studyproposed potential ways to increase prediction quality. The conducted sensitivityanalysis revealed interesting mechanics of SWAT such as sensitivities varying overtime and space and a clear dominance of hydrology related input parameters. Thisstudy concludes that SWAT offers a high potential to analyze nutrient flows andtransformations as well as the spatial pattern of biogeochemical cycling within theZRB. Nevertheless, the fact that ZRB1 did not yield reliable prediction qualitymade it not possible to draw solid conclusions on catchment wide biogeochemistry.Proposed next steps are therefore the quality check of input data, the use of a dif-ferent method to calculate surface runoff and the change of hydrological calibrationroutines followed by a chemical calibration of the model.

The widespread use of pesticides in the environment represents a threat to aquatic organisms. Unfortunately little is known about the actual negative impact of pesticides on aquatic life. A deeper understanding of the processes governing these adverse effects is needed for the definition of water quality criteria and improvement of risk assessment. Toxicokinetics describe the time course of contaminant concentrations in organisms including uptake, elimination and metabolism and are therefore important to understand toxicological processes.<br /> The aim of this study was to assess the time dependent uptake and elimination as well as the formation of metabolites of diazinon in the freshwater invertebrate <em>Gammarus pulex</em>. The acute toxicity of diazinon to <em>Gammarus pulex</em> was determined for a meaningful design of the toxicokinetic tests. Diazinon is a lipophilic compound and will accumulate in<em> Gammarus pulex</em> preferentially in the lipid tissue. For a comprehensive toxicokinetic analysis, the lipid content of <em>Gammarus pulex</em> needs to be determined. There was no lipid determination method for aquatic invertebrate established at EAWAG. Therefore one of the tasks of this thesis was to develop a method for the lipid determination<br /> The acute toxicity of diazinon to <em>Gammarus pulex</em> was determined in short-term mortality assays. The 24hr LC₅₀ (lethal concentration for 50% of test organisms) was 46.6 μg diazinon/l; the 48hr LC₅₀ is 13.1 μg diazinon/l (nominal concentration).<br /> The uptake and elimination rate constants were determined using the computer program OpenModel and were 283 ± 23 (SD)[L kg^-1day^-1] and 3.4 ± 0.3 (SD)[day^-1]. In order to be able to quantify the concentration in the water and in the organism before and after exposure, ¹⁴C radiolabelled diazinon was used. Concentrations were measured through liquid scintillation counting. The metabolites produced in <em>Gammarus pulex</em> after 24 hr exposure at the nominal water concentrations of 5 μg/l and 10 μg/l of ¹⁴C radiolabelled diazinon were measured with the radio-HPLC. Independently of the exposure concentration a fast biotransformation of the parent compound could be observed. At the lower concentration no parent compound could be measured in the organism, at the higher exposure concentration 16% of the parent compound was found. Only one metabolite could be observed at both exposure concentrations. This is most likely pyrimidinol (84%).<br /> Two different methods for the determination of the lipid content were developed: a gravimetric method and a method based on the quantification of the total organic carbon (TOC) and total phosphor. The <em>Gammarus pulex</em> lipid content found was of 5.9 ± 0.1 (% dry weight, ± SD) with the gravimetric method; and 5.4 ± 0.5 (% dry weight, ± SD) with the TOC and total phosphor method. This method allows the distinction of the lipid in two classes: triglycerides and phospholipids.<br /> The amount of triglycerides represented 75.2 ± 9.7 (% tot lipid ± SD ); the phospholipids represented 24.8 ± 9.7 (% tot lipid ± SD).

The Lake Victoria cichlid fish species flock presents a text book example of adaptive radiation and spectacular diversity in terms of morphology, ecology and behaviour. However, this lake which is a home to hundreds of haplochromine species has been subjected to strong loss of diversity during the end of the previous century due to various anthropogenic activities. Combining these two facts, Lake Victoria is an ideal model ecosystem for the study of adaptive radiation, and interactions between environmental heterogeneity, species diversity and biodiversity dynamics in an evolutionary and conservation biology frame work. To this end, this study uses a comparative genome scan to evaluate the impact of ecological gradients on genomic differentiation between replicate pairs of divergent rock dwelling Neochromis spp. from 3 islands in the southern region of Lake Victoria.<br /> We tested how genomic differentiation varies with steepness of ecological gradient using population of Neochromis spp. that live along environmental gradients created by water depth and associated light condition. We hypothesized that 1. Variation in gradient steepness induced by variation in shore slope, water clarity and rock size explains variation between islands in the extent of trophic adaptation and genetic differentiation 2) shallower environmental gradients are more prone to induce genetic differentiation and speciation than steeper environmental gradients.<br /> From our analysis based on 574 polymorphic loci, we observed low to moderate genetic diversity within population. A mean gene diversity (Hex) of 0.11±0.01 and a mean proportion of polymorphic loci varying from 20 to 32% were observed. We found very low genetic differentiation between trophic morphs (FST= 0.4%) at the island with very steep ecological gradients and high genetic differentiation (FST ~4%) at the island with an ecological gradient of intermediate steepness. Furthermore, we found the strongest adaptive differentiation (revealed by highest number of outliers, highest BayeScan over all FST value and Phenotype divergence) in the sympatric trophic morph at the shallow ecological gradients as predicted by theory.<br /> In conclusion we found genomic signature of divergent selection between trophic morphs at Igombe and Makobe revealed by high FST value and strong association of markers to adaptive phenotypic traits. But we did found no evidence of selection at Bihiru Island, with very steep ecological gradients. Thus, our findings support the hypothesis that variation in steepness of resource gradients determines the extent of trophic adaptation and its genomic signature and suggests that shallow resources gradients induce stronger adaptive genetic differentiation compared to steeper resource gradients.

<em>Pundamilia pundamilia</em> and <em>Pundamilia nyererei </em>are very closely related Lake Victoria cichlid species that occupy different microhabitats in rocky habitats but vary in male nuptial colour, a basis for divergent natural and sexual selection. Genomic regions under divergent selection, the so-called “outlier loci” exhibit elevated differentiation (measured as F_ST) and can be detected by a genome scan using a large number of genomic markers. We used, 516 AFLP markers fiom 4 paired phenotype populations collected from 4 Islands, that form “the <em>Pundamilia </em>speciation transect” in the southern part of Lake Victoria varying in water turbidity and steepness of light slope. Genetic differentiation between the sympatric colour morphs was highly significant (p&lt;0.001***) except at the island with highest turbidity (Luanso; p = 0.069), and decreased with decrease in water transparency. A highly significant differentiation was found between islands in pooled sympatric morphs (F_CT = 0.041; p&lt;0.001**) but not in pooled populations of each morph across islands (F_CT=-0.011; p = 0.8). Of the 516 AFLP loci, 3 outlier loci (4N287, 4B348 and 2Nl86) at Makobe and 1 outlier locus (2G225) at Python were identified using a stringent detection threshold in Bayescan. No outliers were detected at the more turbid Islands of Kissenda and Luanso. Relaxing the detection threshold from a logl0 Bayes factor of 2 (<em>p</em>&lt;0.01) to 1 (<em>p</em>

The rapid development of nanotechnology and the widespread production and use of nanomaterials will inevitably lead to their appearance in the environment. On the basis of the use of silver nanoparticles (AgNP) in several consumer products, it is likely that AgNP will be released to the aquatic environment. To evaluate the potential environmental impacts associated with AgNP, dissolution of AgNP and their toxicity to the freshwater green algae <em>Chlamydomonas reinhardtii</em> were examined.<br /> The dissolution of Ag⁺ from AgNP was measured using a silver ion-sensitive electrode (ISE). Reproducible linear Nernstian response of ISE could be achieved in a logarithmic Ag⁺ concentration range between -6 and -3. To increase its sensitivity ligands which complex Ag⁺ have been added. Using bromide and chloride as silver ligands the constructed calibration curves with a linear Nernstian response of the electrode could be extended until free log [Ag⁺] of around -10 and -8, respectively. Ag⁺ measurements using Ag-ISE in diluted AgNP suspensions between 92.7 and 927 μM found dissolution between 0.88 and 1.37 % of the total silver in AgNP. Considering that entering the aquatic environment AgNP may be exposed to various pHs, the effect of pH on the dissolution of AgNP were assessed in short term experiments (&lt; 3 h). Dissolution of AgNP differed slightly exposing AgNP at pH 6, 7 and 8; and a decrease of the measured Ag⁺ concentrations over time was observed.<br /> The toxicity of AgNP to <em>Chlamydomonas reinhardtii </em>was measured as the reduction in photosynthetic activity using fluorometry. A time- and concentration- dependent photosynthesis inhibition of <em>Chlamydomonas reinhardtii</em> exposed to AgNP has been observed. In experiments exposing algae to two different AgNP concentrations and in presence of various chloride concentrations the role of Ag⁺ in explaining AgNP toxicity was evidenced. Comparing EC₅₀ values expressed as function of Ag⁺ and calculated by exposing <em>Chlamydomonas reinhardtii</em> to Ag⁺ and AgNP showed that toxicity of AgNP is higher than that of Ag⁺. This may indicate an additional Ag⁺ formation upon AgNP-cell contact.<br /><br /> Die rasanten Entwicklungen im Bereich der Nanotechnologie in den letzten Jahren, führte zum Vorhandensein vieler neuer Produkte, die Nanopartikel enthalten. Silbernanopartikel (AgNP) finden in zahlreichen Konsumentenprodukten Anwendung. Während der Nutzung können sich AgNP heraus lösen und in die Gewässer gelangen. Um die potentiellen Effekte von AgNP auf die aquatische Umwelt zu untersuchen, wurde die Auflösung von AgNP sowie deren Toxizität auf Süsswasser Grünalgen (<em>Chlamydomonas reinhardtii</em>) untersucht.<br /> Mit einer ionen-selektiven Elektrode wurde die Auflösung freier Silberionen (Ag⁺) aus AgNP gemessen. Im Bereich einer logarithmischen Ag⁺-Konzentration von -6 M bis -3 M konnte mit der ISE eine reproduzierbare lineare Nernstsche Antwort erzielt werden. Um die Sensitivität der ISE zu erhöhen, wurden Liganden zugegeben, die Ag⁺ komplexieren. Dank den Liganden (Bromid bzw. Chlorid) konnte die lineare Nernstsche Antwort auf noch geringere -Konzentrationen (log [Ag⁺] von -10 M bzw. -8 M) angewandt werden. Die Ag-ISE-Messungen ergaben, dass sich aus verdünnten Suspension die 92.7 bzw. 927 μM AgNP enthalten, 0.88 bzw. 1.37% freie Silberionen lösen. In aquatischen Systemen können AgNP unterschiedlichen pH-Werten ausgesetzt sein. Im Rahmen von Kurzzeitexperimenten (&lt; 3h) wurde die Auflösung von AgNP bei unterschiedlichen pH-Werten analysiert. Im Bereich von pH 6, 7, 8 variiert die Auflösung von AgNP nur geringfügig. Die Ag⁺-Konzentration sank jedoch im Laufe der Zeit.<br /> Eine Reduktion der photosynthetischen Aktivität bei <em>Chlamydomonas reinhardtii</em> galt als Indiz für die Toxizität von AgNP und wurde mittels eines Fluorometers gemessen. <em>Chlamydomonas reinhardtii</em>, die AgNP ausgesetzt waren, zeigten eine zeit- und konzentrationsabhängige Hemmung der Photosynthese. Die Rolle von Ag⁺ im Rahmen der AgNP-Toxizität wurde deutlich, als die Algen in Gegenwart variierender Ligandenkonzentrationen (Chlorid) zwei unterschiedlichen AgNP-Konzentrationen ausgesetzt wurden. Im Weiteren wurden die Algen separat einerseits Ag⁺-Ionen und andererseits AgNP ausgesetzt. Als Funktion der Ag⁺-Konzentration wurden daraus EC₅₀ Werte berechnet. Ein Vergleich der EC₅₀ Werte zeigt, dass die Toxizität von AgNP grösser ist, als die von Ag⁺. Dies könnte darauf hindeuten, dass der Kontakt von AgNP mit der Zelle zur Auflösung weiterer Ag⁺ führt.

In alpinen Regionen stellen Wildbäche immer wieder ein beträchtliches Schadenspotential dar. Bei intensiven Niederschlägen können deren Abflüsse rasch stark ansteigen, was nicht nur zu Überflutungen führt, sondern auch zum Transport von Geschiebe. Gerade in sehr dicht besiedelten Gebieten wie der Schweiz befinden sich viele Gebäude, Infrastruktur und andere Werte in Reichweite solcher Hochwasser- und Geschiebetransportereignisse. Daher ist es wichtig, das Ausmass der Gefahren prognostizieren zu können, um im Sinne eines integralen Risikomanagements entweder betroffene Gebiete unbebaut zu lassen oder diese Zonen zu schützen. Zur Abschätzung der von Wildbächen während Hochwasser geführten Geschiebefrachten wurde das Simulationsprogramm <em>SETRAC</em> (<em>SE</em>diment <em>TR</em>ansport in <em>A</em>lpine <em>C</em>atchments) entwickelt, mit welchem in der vorliegenden Studie modelliert wurde.<br /> Ausgangslage der Studie sind die Hochwasser vom August 2005, die fast die gesamte Alpennordseite der Schweiz betrafen und Schäden in einer Höhe von knapp 3 Milliarden Schweizer Franken verursachten. Zahlreiche Wildbäche mobilisierten enorme Mengen an Geschiebe. In dieser Arbeit wurden zwei dieser Bäche ausgewählt und näher untersucht: der Buoholzbach und der Steinibach Dallenwil im Kanton Nidwalden. Für beide Bäche wurden nach den Hochwasserereignissen 2005 die Geschiebefrachten im Feld abgeschätzt. Zusätzlich wurden für diese Arbeit Geschiebebilanzen aufgestellt, die gemeinsam mit der Frachtabschätzung genutzt wurden, um die Güte der Simulationsergebnisse von <em>SETRAC</em> zu bewerten. Ein Grossteil der Gerinnestrukturen und weitere Kenngrössen der Einzugsgebiete wurden im Rahmen dieser Studie im Feld gemessen. Die zur Modellierung notwendigen Abflussganglinien wurden mittels Niederschlag-Abfluss-Simulation bestimmt.<br /> Ziel der Studie war es, die in <em>SETRAC</em> implementierten Ansätze zur Berechnung von Fliesswiderstand, Geschiebetransport, Bewegungsbeginn des Sediments und zur Berücksichtigung der Formverluste zu plausibilisieren. Dazu wurden pro Bach je 18 unterschiedliche Simulationen laufen gelassen. Die Interpretation der Simulationsergebnisse ist schwierig, da die zur Kalibrierung benutzten beobachteten und rückgerechneten Geschiebefrachten grosse Unsicherheiten aufweisen. Dennoch stimmen für den Steinibach in 12 von 18 Fällen und für den Buoholzbach in 11 von 18 Fällen die errechneten mit den beobachteten Geschiebefrachten überein. Für den Buoholzbach wurden die Frachten aber tendenziell unterschätzt und 3 Simulationsrechnungen führten zu unrealistischen Ergebnissen, was auf numerische Probleme in <em>SETRAC</em> zurückzuführen war. Aufgrund ausufernder Rechenzeiten konnten für den Buoholzbach 5 Simulationen und für den Steinibach eine Simulation nicht zu Ende gerechnet werden.<br /> Mit dieser Arbeit konnte gezeigt werden, dass ein realistisches Ergebnis für die Geschiebefrachten stark von einer geeigneten Wahl der in <em>SETRAC</em> implementierten Ansätze abhängt. Die von anderen Autoren vorgeschlagenen, optimalen Gleichungskombinationen konnten bestätigt werden. Die Analyse der Ergebnisse zeigte, dass die Relevanz der untersuchten Faktoren bezüglich den abgeschätzten Geschiebefrachten in dieser Reihenfolge zunimmt: die Wahl des Formverlust-Exponenten , die Wahl des Ansatzes für die Formverlust-Korrektur und die Wahl des Ansatzes für den Bewegungsbeginn des Geschiebetransportes. Die Ergebnisse, die mittels der verschiedenen Ansätze zur Berechnung von Bewegungsbeginn und Formverlusten errechnet wurden, weichen um einen Faktor 10 und mehr voneinander ab.

Lake Kariba and Lake Itezhi‐Tezhi are two man‐made hydropower reservoirs located in the Zambezi River Basin in southern central Africa. Different studies have shown that these reservoirs have negative influence on downstream ecosystems. The goal of this study was to describe the biogeochemical properties of these reservoirs by analysis of sediment records, to get a better understanding of the impact of these reservoirs on downstream transport of carbon and nutrients. This study thereby focused on differences in the proportion of terrestrial and lacustrine organic matter, spatial variation in the reservoirs, historic changes and trends and the potential of the reservoirs to trap particles behind their walls.<br /> Visual, physical and chemical analysis of samples from sediment cores collected along transects from the two main inflows to the dams revealed high spatial variation in the biogeochemistry of oth reservoirs. For most of the analyzed parameters, gradients from the inflows to the dam were found:<br /> • Light beige colored se diments change to dark colored sediments.<br /> • Sediments dominated by quartz minerals with particles of the coarse silt to fine sand fraction are replaced by sediments dominated by organic matter and clay minerals in the clay to coarse silt fraction.<br /> • Organic carbon content increases from 2 – 3% to 3 – 5% (Kariba) and from 3 – 5% to 4 – 7%(Itezhi‐Tezhi), similar to an increase of total nitrogen and total phosphorous concentrations.<br /> • Atomic C:N ratios decrease from 16 – 18 to 10 – 14 (Kariba) and 12 – 13 to 11 – 12 (Itezhi‐Tezhi).<br /> • Carbon isotope ratios in Lake Kariba decrease from around ‐20‰ to values ranging from ‐27‰ to ‐24‰, while they do not change significantly in Lake Itezhi‐Tezhi and range from ‐25‰ to ‐22‰. Nitrogen isotope ratios do not change either in both reservoirs and range from 0‰ to 5‰.<br /> Overall, these trends indicate the importance of terrestrial organic matter settling near the inflows, in opposition to a higher portion of autochthonous organic matter being deposited closer to the dams. In addition to spatial differences high temporal variation was detected, supposedly caused by the event‐driven character of the sedimentation regime in both systems. The cores collected far from the inflows show irregularly distributed layers with very different properties. These differences derive from the temporal influence of floods and autochthonous production. Flood layers contain a higher proportion of terrestrial organic matter, their organic matter content is lower and they consist of coarser material. The flood layers therefore resemble the sediments found near the inflows. The differences between layers are smaller in Lake Itezhi‐Tezhi because 2 the terrestrial influence on sediment properties is generally higher in this reservoir, due to its smaller size.<br /> Some of the cores comprise two halves with pre‐dam sediments below sediments deposited after dam closure. The transition zone between the two halves features abrupt changes in bulk density, carbon, nitrogen and phosphorus concentrations, C:N and carbon isotope ratios. Hence, these changes could roughly be dated to the dam closures (~1960 for Kariba, ~1977 for Itezhi‐Tezhi). Although all cores could not be dated using radio nuclides, for cores containing pre‐dam sediments average sedimentation rates and mass accumulation rates of solid matter, carbon, nitrogen and phosphorus could be calculated. For the remaining cores, only minimum values could be stated. In summary, sedimentation rates in Lake Kariba decrease from more than 0.56 – 0.70 cm/yr to 0.19 – 0.50 cm/yr along transects from inflows to the dam. Sedimentation rates in Lake Itezhi-Tezhi range from 0.28 cm/yr (at the dam) to more than 1.9 cm/yr (minimum value close to the major inflow). The data suggests that in both reservoirs a significant proportion of material is deposited near the river mouths. Using these estimated sedimentation rates and the resulting mass accumulation rates, the accumulated mass of solid matter, carbon, nitrogen, and phosphorous were calculated. The resulting values could serve as sink terms in preliminary mass balances, which will help in quantifying the trapping potential of the reservoirs.

Important amounts of nutrients excreted by humans are found in human urine. This provides the motivation for separating urine and recycling it, as fertilizer, back to agricultural land for food production. Urine diverting toilets have been increasingly being promoted in Nepal and more than 500 units have been installed since 2002 (Das and Kumar, 2008). However, potential drawbacks to urine handling systems are the risk of ammonia evaporation and the relatively large volumes to be handled. In that situation struvite technology has been established to trap the phosphates in a solid fertilizer. Using the technology could reduce the huge volume of urine and transporting cost. However, struvite production also generates effluent. Struvite effluent reuse has significant potential benefits on both a local and global scale, such as re-circulating plant nutrients like nitrogen and potassium back to agriculture.<br /> This thesis explores the potential reuse of struvite effluent in Siddhipur Nepal. The hypotheses being investigated are that 1) preliminary struvite precipitation prevents clogging during drip fertigation with urine and 2) that drip-fertigation with urine is superior to bucket spreading, because the ammonia volatilization is strongly reduced.<br /> When the nutrients in effluent are beneficially utilized through irrigation some amount of pollutants discharged into our waterways can be reduced.<br /> In this study the small plots for drip irrigation and ammonia volatilization were established. The amount of clogging was based on measurements of head loss in the tank and time of application. The ammonia volatilization measurements were collected in each hour after application.<br /> Boric acid was used to trap the ammonia volatilized in the plots. The method was successfully used to determine that more ammonia volatilized in the plot of the bucket application for up to 2.3 times as compared with drip effluent plot. The largest loss (11% of the applied N) was measured after application of 45 L of urine per plot. Virtually no volatilizations were detected (on the first 6hrs) when the urine was applied to the freshly tilled soil. The results also suggest that reuse of struvite effluent through drip irrigation is possible without full clogging (only 30% decrease of flow rate).<br /> The reuse of effluent from a struvite reactor is beneficial in terms of the nutrients that are made available and when applied through drip irrigation the loss of ammonia is reduced. In addition, the reuse of effluent can contribute towards the reduction of importing the commercial fertilizers.

In der Veterinärmedizin eingesetzte Sulfonamid-Antibiotika, die mit der Gülle in landwirtschaftlich genutzte Böden eingetragen werden, könnten dort die Antibiotikaresistenz von Bakterien induzieren und die natürliche Mikroorganismenpopulation negativ beeinflussen. Ein entscheidender Faktor dafür ist die Bioverfügbarkeit der Sulfonamide. Sorption und Desorption sind zwei wichtige Prozesse, die Bioverfügbarkeit sowie auch Abbau und Transport massgeblich bestimmen. Der wichtigste Sorbent für Sulfonamide im Boden ist die organische Substanz. Das Ziel dieser Arbeit war die es, die Desorbierbarkeit bzw. Extrahierbarkeit von Sulfonamiden von Huminsäure zu untersuchen. Dafür wurde Sulfathiazol (4-Amino-N-(2-thiazolyl)-Benzensulfonamid, STZ) und 4-Methyl-N-(2-thiazolyl)-Benzensulfonamid (MTBS) an (I) gelöste, (II) mit Aluminium aggregierte und (III) Laccase-inkubierte Leonardite-Huminsäure im Batchversuch sorbiert. Die Desorption von der Huminsäurelösung (Halbzelle 1) erfolgte mit einem Festphasenextraktionssorbenten (Halbzelle 2) in einem Zell-Zell-System, welches durch eine Dialysemembran getrennt war. [...]<br /><br /> Veterinary sulfonamide antibiotics that reach agricultural soils via liquid manure might induce the formation of antibiotic resistance genes and negatively affect the natural population of microorganisms. An important factor for the risk assessment is the knowledge of the bioavailability of the sulfonamides. Sorption and desorption are two essential processes influencing bioavailability as well as degradation and transport. Organic matter is the most important sorbent for sulfonamides in soil. The goal of this work is to analyze the desorbability and extractability of sulfonamides from humic acid. To this end sorption experiments were conducted with sulfathiazole (4-amino-N-(2-thiazolyl)-benzene-sulfonamide, STZ) and 4-methyl-N-(2-thiazolyl)-benzene sulfonamide (MTBS) with (I) dissolved, (II) aluminium-aggregated and (III) laccaseincubated Leonardite humic acid in batch experiments. While the aniline group of STZ might covalently bind to the organic matter, MTBS possesses an unreactive tolyl group. Fast runoff from agricultural areas contributes to diffuse pollution of surface water bodies. Generally, the fast runoff originates only from a limited part of the catchment. The identification of this part relies on spatially distributed models which predict the hydrological processes. I address the question whether simple models are capable to represent the relevant runoff processes. I compare three spatially distributed models of different complexity: the FAL risk assessment of nitrogen and phosphorus losses, an approach to delineate the dominant hydrological runoff processes (DRP) and the Soil Moisture Distribution and Routing model (SMDR). The former two models are expert systems, whereas the latter is a mechanistic model. These three models were applied to a small agricultural catchment in the Swiss Plateau. The comparison reveals that the models agree in the general spatial predictions of runoff patterns. A low FAL risk corresponds to a slow reacting runoff process based on DRP and a low vulnerability to runoff formation based on SMDR. But the SMDR predictions vary to a large extent within the FAL and DRP classes because SMDR takes interactions between adjacent areas into account. Such spatial interactions are neglected by the two expert systems. In a second part of my thesis, I attempt to improve the simple expert-type models by deriving a meta-model from the output of the mechanistic SMDR simulations. A regression tree was built which explains the surface runoff pattern in the study area as predicted by SMDR. The tree resembles the spatial distribution fairly well. Nevertheless, it only explains 52 % of the variability. The limited amount of explained variability suggests that a new measure needs to be developed to better represent the interactions between adjacent areas. Such a measure should consider the topography and the soil properties within the specific catchment of an area.

Im Rahmen des Vision of Future Energy Networks (VoFEN) Projekts am Institut für elektrische Energieübertragung und Hochspannung der ETH Zürich wurde das Multi-Energie-Portfoliomodell entwickelt. Das Modell ermöglicht, effiziente Erzeugungsportfolios für die Bereitstellung mehrerer Energieträger zu bestimmen. Die vorliegende Arbeit wendet das Multi-Energie-Portfoliomodell als Fallbeispiel auf das zukünftige Strom- und Wärmeportfolio der Stadt Zürich an. Unter der Berücksichtigung der zwei Energieszenarien ‘2000-Watt-Gesellschaft’ und ‘realistisch-ambitiös’ der Stadt Zürich werden für die Zeitpunkte 2030/40 und 2050/60 effiziente Erzeugungsportfolios berechnet. Die Optimierung der Portfolios basiert auf dem erwarteten Portfolioertrag und der Standardabweichung dieses Ertrags als Mass für das Risiko. Zusätzlich enthält das Modell Nebenbedingungen, um die spezifischen Anforderungen des Fallbeispiels berücksichtigen zu können. Mit den Nebenbedingungen wird das Strom - Wärme Verhältnis des Portfolios festgelegt, die technischen Potenziale der Erzeugungstechnologien begrenzt sowie die CO₂äq Emissionen und der Primärenergieverbrauch des Portfolios limitiert. Die Angaben zu den erwarteten Gestehungskosten für die unterschiedlichen Erzeugungstechnologien stammen aus mehreren Studien. Im Modell ist der Ertrag einer Technologie als das Inverse deren Gestehungskosten definiert. Insgesamt werden 26 verschiedene Technologien modelliert. Die Resultate zeigen, dass alle effizienten Portfolios die Wärmeerzeugungstechnologien Solarkollektoren, Wärmepumpen und Biomasse (Heizung) enthalten. Strom und Wärme erzeugt mit der Wärmekraftkopplungstechnologie Dampfturbine KVA sind Bestandteile aller effizienten Portfolios. Die Anteile der weiteren Technologien an den Portfolios sind vom Zeitpunkt, den Nebenbedingungen und dem Portfoliorisiko abhängig. Fossile Grosskraftwerke, die mit Gas oder Kohle gefeuert werden und keine CO₂ Abscheidung und Speicherung haben, kommen in keinem der modellierten zukünftigen Portfolios vor. Wie die vorliegende Arbeit aufzeigt, kann das erweiterte Multi-Energie-Portfoliomodell<br /> als Instrument zur Ermittlung zukünftiger effizienter Erzeugungsportfolios verwendet werden.<br /><br /> The Multi-Energy Generation Portfolio Model has been developed within the research project Vision of Future Energy Networks (VoFEN) at the Power Systems and High Voltage Laboratories at the ETH Zurich. This model enables to ascertain efficient portfolios for the production of several energy carriers. As a case study the Multi-Energy Generation Portfolio<br /> Model is used to determine the future electricity and heat portfolio of the city of Zurich. Efficient Portfolios are calculated considering the two energy scenarios ‘2000 Watt-Society’ and ‘realistic-ambitious’ for the dates 2030/40 and 2050/60. The optimization is based on the expected return of the portfolio and its standard deviation which is considered as a measure for the risk of holding the portfolio. Additionally the model includes constraints<br /> which enable to account for specific demands of the case study city. The constraints define the ratio of electricity and heat produced by the portfolio and limit the technical potential of the technologies as well as the greenhouse gas emissions and the use of primary energy by the portfolio. Several studies have been considered to determine the expected production costs of the energy technologies in future. The return of a technology is defined as the inverse of its production costs. In total 26 different technologies are modelled. All calculated efficient portfolios contain the heat technologies solar collector, heat pumps, and biomass (for heating). Combined heat and power generation based on the combustion of municipal solid waste is also part of all efficient portfolios. The shares of the other technologies in the portfolio depend on the dates, the constraints, and the portfolio risk. Fossil fuel fired large-scale power plants without carbon capture and storage are not part of any modelled efficient portfolio. As shown within this work the extended Multi-Energy Generation Portfolio Model allows to model efficient energy portfolios for the future.

Struvite (MgNH₄PO₄·6H₂O) recovery from wastewater streams has been known as an efficient nutrient recycling method. However, current research has predominantly addressed high-end solutions to overcome fertilizer shortages in agriculture and prevent eutrophication in receiving waters.<br /> The present study focused on low-cost struvite recovery from stored human urine, by optimizing a process developed at pilot scale in the peri-urban settlement of Siddhipur in the Kathmandu Valley (Nepal). A large amount of world’s species diversity is the result of adaptive diversification to heterogeneous environments. Such species are reproductively isolated from each other by ecologically-based post zygotic isolation or mate choice. Changing environments can cause the breakdown of genetic isolation between such species. It has been shown that a loss of environmental heterogeneity can lead to fusion of species, on the other hand interspecific hybridization can under some circumstance contribute to increase biodiversity. To measure extent of genetic differentiation between two sympatric whitefish species of Lake Lucerne, we performed a quantitative sampling along an ecological gradient. Instead of the previously described known two species we found three genetically differentiated species along the gradient that further differ in morphology and life-history traits. The newly discovered species is genetically intermediate between the two previously known species but distinct from both. We discuss alternative hypothesis for its origin including the possibility that the recent environmental changes led to hybrid speciation and we discuss the effect of changing environmental conditions on the interaction between gene flow and selection and on the conservation of species.

Preliminary studies have shown that source-separated urine is a suitable medium for nitrogen removal via aerobic and anoxic ammonia oxidation (Udert et al., 2003). Furthermore, denitrifying bacteria can use the sufficient high concentration of dissolved organic matter, which is build up during the anammox process. Combination of all aerobic and anoxic processes leads to an increased nitrogen removal and savings in the oxygen consumption. On the basis of previous studies, a single aerobic/anoxic sequencing batch reactor was chosen as a most powerful tool for the integration of these microbiological processes.<br /> During this diploma thesis further experiments were made in order to better understand the overall process of nitrogen removal. Thanks to intensive sampling some problems in the course of the reactor were observed. It was noticed that at the beginning of each cycle, only organic matter is consumed, whereas nitrogen concentration stays at the same level. This caused a delay in the nitrogen elimination and was considered as a dysfunction of the process. The evaluation of the toxicity of urine showed that urine contains some toxic substances which are responsible for the inhibition of ammonium oxidizing bacteria. However, it could be observed that during the course of the cycle, the toxicity decreases to a lower level which enables autotrophic bacteria to start ammonium oxidation. Since during this initial time only heterotrophic bacteria show their activity, the decline of toxicity was attributed to these microorganisms. Thanks to this observation it could be showed that heterotrophs play a very important role in the overall process: on the one side through anoxic respiration they reduce nitrite and nitrate which results in better overall nitrogen removal, and on the other side via removal of organic matter they decrease the inhibition effects on ammonium oxidizing bacteria.<br /><br /> Bisherige Studien zeigten, dass ein einstufiges Nitritation-Anammox-Verfahren eine effiziente Lösung zur Stickstoffentfernung ist. Udert et al. (2003) konnten beweißen, dass das Verfahren sich ebenfalls für die Behandlung von menschlichen Urin gut eignet. Urin enthält große Mengen von Ammonium und relativ hohe Konzentrationen organischer Substanz. Diese Zusammensetzung ermöglicht neben einer autotrophen Stickstoffentfernung (Ammonium-Oxidation folgend durch Anammox) auch eine heterotrophe (Denitrifikation) Stickstoffentfernung. Eine Kombination dieser mikrobiologischen Prozesse erhöht die Effizienz und reduziert die Kosten des Verfahrens. Zum optimalen Einsatz des Verfahrens wurde SBR Reaktor gewählt.<br /> Im Rahmen dieser Diplomarbeit wurden weitere Versuche durchgeführt, die dem besseren Verständnis und der Charakterisierung der Ablaufenden mikrobiologischen Prozesse dienten. Anhand von gemessenen Daten, konnte festgestellt werden, dass die Stickstoffentfernung gestört ist. Beobachtungen zeigten, dass am Anfang des SBR-Zyklus nur die organische Substanz assimiliert wird und keine Ammonium-Oxidation stattfindet. Demzufolge wird die Stickstoffentfernung in der Zeit versetzt, was zur Verlängerung des Zyklus führt. Weitergehende Untersuchungen zeigten, dass Urin toxische Stoffe enthält, die für diese Inhibierung der Ammonium-Oxidanten verantwortlich sein könnten. Es wurde beobachtet, dass während eines SBR-Zyklus die gemessene Toxizität sinkt: demzufolge nehmen auch die Inhibierungseffekte auf die autotrophen Mikroorganismen ab und die Ammonium-Oxidation kann beginnen. Diese Abnahme der Toxizität geschieht in der ersten Phase des Zyklus. In dieser Zeit sind die heterotrophen Bakterien am aktivsten. Daher wirkt sich die heterotrophe Aktivität auf den gesamten Prozess positiv aus.<br /> Als Resümee der Versuche, stellte sich heraus, dass die heterotrophen Mikroorganismen eine große Rolle in dem Verfahren spielen. Einerseits werden durch die anoxische Atmung Nitrit und Nitrat entfernt, was zur Effizienz des Prozesses beiträgt. Andererseits werden durch ihre aerobe Aktivität die toxischen Stoffe abgebaut und die Ammonium-Oxidation ermöglicht.

The enzyme LinB belongs to the enzyme family of haloalkane dehalogenases. This class is very interesting for biochemical work due to its ability to transform halogenated compounds to the corresponding alcohol without requiring any cofactor or cosubstrate except water. These enzymes are also interesting for the chemical or pharmaceutical industry to use them for the synthesis of chiral intermediates or recycling of chlorinated byproducts from chemical production.<br /> In this work LinB enzymes from two different sources were characterized regarding their substrate specificities and possible enantioselective reactions towards chlorinated and/or brominated racemic substrates. The following results were obtained.<br /> 1. Different expression systems were tested to produce recombinant LinB from S. indicum B90A and <em>S. japonicum</em> UT26 in <em>E. coli</em>. The highest activity in the crude cell extract was reached with LinB from B90A that carried an N-terminal His-tag.<br /> 2. For measuring the LinB activity an analytical method was needed. A standard assay for the measurement of LinB activity by analytical ion chromatography was developed.<br /> 3. One enzyme variant, His-LinB B90A, was purified by Ni-NTA chromatography to analyze the substrate specificity.<br /> 4. Nine chlorinated and/or brominated substrates were tested and the specific activities ranged from 0.14 to 1.85 U/mg of protein. 3-Chloro2- methylpropene showed the highest activity.<br /> 5. The enantioselectivity of LinB was analyzed by NMR with the substrate 2- bromobutane.

Membrane aerated bioreactors (MABR) provide attachment for biofilm and supply air for biological processes.<br /> An MABR operated in coninously stirred tank reactor (CSTR) treats undiluted urine (high ammonium, pH value around 9) to produce NH₄-NO₃ with a pH value around 6. The main influence factors investigated in the system were mainly the free ammonia (FA), free nitrous acid (FNA) and carbonate concentration.<br /> The experiment was designed into 3 phases. Phase 1 was operated in batch-mode using tapwater as start-up medium. Phases 2 and 3 were started with a conditioning saline solution and then switched to the CSTR mode. Activated sludge was used as inoculum for biofilm in phase 1. Sludge used to build conditioned biofilm in phase 2 was a mixture of equal volumes of activated sludge and sludge from a partial nitratation and anammox (PNAA) system. In phase 3, the biofilm was grown from activated sludge and the NOB of the biofilm were selectively cultured using 10ml batch doses of 0.5M NaNO2 .<br /> Influent paramenters analysed were NH₄, NO₂, NO₃, dissolved COD, total inorganic carbon (TIC) whilst in the effluent, the parameters determined were NH₄, NO₂, NO₃ and TIC. Dissolved oxygen (DO) concentration and pH in the reactor were taken daily.<br /> During phase 1, COD degradation and ammonium loss were observed. Nitrification was inhibited by FA concentration of above 300mg/l though the value later decreased to 40mg/l. Average oxygen concentration was 9mg/l and pH of the medium averaged above 8.0. Effect of FNA was not observed. During phase 2, a constant load of 0.12gN/d was supplied. FA concentration gradually increased to 40mg/l and eventually inhibited NOB activity but the AOB continuously produced nitrite. There was no FNA effect. Average oxygen concentration was 8mg/l and pH ranged between 7 and 8. In phase 3, varying loads of 0.05 to 0.22gN/d was delivered. Maximum concentration of FA was 0.07mg/l and the highest value of FNA was 0. 09mg/l. Average oxygen concentration was 7.0mg/l and pH averaged 6.5. There was hardly measurable carbonate in the system though NOB and AOB continuously functioned that a solution of ammonium nitrate (1:3) was produced at non-steady state.

In this study, the modes of operation of prompts and public commitment were investigated in order to gain knowledge about how these two intervention techniques induce behavior change. For this purpose a theoretical model of how the interventions operate was established and tested with path analyses on longitudinal data from a field study in Bolivia. The field study was about changing people’s behavior in disinfecting their drinking water with SODIS (solar water disinfection). Within the models, four behavior-near constructs were aimed to explain the effects of prompts and public commitment: habit, strength of implementation intention, strength of commitment and alternative behavior. The behavior change models were calculated, integrating data from two surveys (two measurement times). It was hypothesized that the four psychological variables affect the behavior before and after the implementation of the interventions.<br /> The results of the path analyses show that a behavior change before an intervention is only influenced by habit and alternative behavior. The prompt affected habit, strength of implementation intention and strength of commitment, though the latter was the only variable that did not show an influence on behavior. However, the path model containing the mode of operation of the public commitment intervention yielded different results. The public commitment operated only on the strength of implementation intention, which strongly influenced the habit and therefore affected the target behavior solely in an indirect way (via habit).

Todays surface waters are exposed to many anthropogenically used compounds (e.g., nutrients, antibiotics, biocides, etc.). Biocides and pesticides belong to some of the most harmful ones, as they are originally designed and used to kill organisms or to render them harmless. Biocides and pesticides are used in urban as well as in agricultural environments. Agricultural pesticide use has so far received much more attention than that of urban biocides. Only recently several studies on the consumption amounts and on surface water exposures indicated that urban sources may be important.<br />The aim of this thesis is to assess simultaneously the importance of urban and agricultural biocide and pesticide use on the compound dynamics in surface waters. Most biocides and pesticides are transported during rain events to surface waters. Hence, this work focuses on the dynamic during rain events throughout the year in a catchment (25 km²) with mixed urban and agricultural land use in the Swiss plateau. [...]<br /><br />Die heutigen Oberflächengewässer sind verschiedenartigsten Belastungen durch anthropogen verwendete Substanzen ausgesetzt (z.B. Nährstoffe, Antibotika, Biozide etc.). Biozide und Pestizide gehören dabei zu den kritischsten Substanzen, da sie zur Schädlingsbekämpfung entwickelt und verwendet werden. Biozide und Pestizide werden sowohl im Siedlungsbereich (urban), als auch in der Landwirtschaft verwendet, wobei bis heute die landwirtschaftlichen Pestizidverluste viel stärker untersucht wurden als diejenigen aus der Siedlung. Erst in den letzten Jahren haben erste Studien gezeigt, dass sich einerseits der Verbrauch in einem ähnlichen Rahmen bewegt, dass aber auch der Biozideintrag in die Gewässer nicht zu vernachlässigen ist.<br />Das Ziel der hier präsentierten Studie ist es, besser zu verstehen, wie die urbanen und landwirtschaftlichen Quellen die Belastung der Oberflächengewässer mit Bio- und Pestiziden beeinflussen. Die meisten Bio- und Pestizide werden während Regenereignissen in die Gewässer transportiert. Deshalb fokkusierte diese Arbeit auf die Schadstoffdynamik während verschiedener Regenereignisse verteilt über ein ganzes Jahr. Das betrachtete Einzugsgebiet (25 km²) hat eine für das Schweizer Mitteland typische, gemischt landwirtschaftlich-urbane Landnutzung. [...]

Lead is a non-essential trace metal, occurring in elevated concentrations in many aquatic systems. Uptake of non-essential trace metals in algae leads to interactions with macromolecules, potentially inducing toxic effects. Algae possess protective mechanisms as a response to metal uptake and toxicity. One of these mechanisms is the control of intracellular metal speciation by synthesis of metal-binding ligands, such as phytochelatins. Phytochelatins (PCs) are metal-binding oligopeptides with the general structure (γ-Glu-Cys)_n-Gly (n = 2 – 11), which are enzymatically synthesized from glutathione. Metal detoxification by PC is assumed to result through immobilization of metals, preventing non-specific binding of metals to important biomolecules, followed by the transport of these complexes into vacuoles. The aim of this project was to examine induction of PC synthesis by Pb in the unicellular freshwater alga <em>Chlamydomonas reinhardtii </em>as a function of the Pb speciation and to explore the role of phytochelatins in Pb detoxification. Furthermore, Pb induced metal-phytochelatin complexes and standard Pb-PC complexes were analyzed to examine whether Pb is actually bound to phytochelatins. Die Aufnahme von nicht essentiellen Spurenmetallen in Algen führt zu Interaktionen mit Makromolekülen und dadurch zu Toxizität. Algen verfügen über Schutzmechanismen als Antwort auf Metallaufnahme und -toxizität. Ein spezifischer Mechanismus ist die Kontrolle der intrazellulären Metallspeziierung durch metallbindende Liganden, zum Beispiel Phytochelatine. Phytochelatine (PCs) sind metallbindende Oligopeptide mit der Struktur (γ-Glu-Cys)_n-Gly (n = 2 – 11), die enzymatisch aus Glutathion synthetisiert werden. Die vermutete Rolle von PC in der Metalldetoxifizierung erfolgt durch Immobilisierung von Metallen, die das nicht-spezifische Binden an Biomoleküle verhindern, gefolgt vom Transport des Komplexes in die Vakuole. Das Ziel dieses Projekts war es die Induktion der PC Synthese durch Pb in der einzelligen Süsswasseralge<em> Chlamydomonas reinhardtii </em>zu erforschen sowie die Rolle von Phytochelatinen in der Pb-Detoxifizierung zu untersuchen. Host susceptibility to parasitism may be strongly influenced by the environment but, importantly, parasite growth is also context dependent. This basic tenet is why natural populations often show genetic variation in host resistance, forming the basis for evolutionary responses to selection induced by parasitism. Hence, biotic factors as well as abiotic environmental variation can affect the expression of traits in hosts and parasites, and thus ultimately alter the fitness of both. In this thesis, I focused on understanding how environmental variation can influence host-parasite coevolution. Different environmental factors can influence hostparasite coevolution; some being more important for host fitness while others constraining parasite fitness more severely. Further, most of these environmental factors are not independent or interact with one another. In this thesis, I intensively examined the influence and potential interactions of four different factors on host-parasite fitness. [...]<br /><br />Die anfälligkeit von Wirten für parasiten kann stark von der Umwelt beeinflusst werden. Zudem hängt das Parasitenwachstum vom allgemeinen Zusammenhang ab. Die zentrale Frage ist daher, warum natürliche Wirtspopulationen häufig genetische Unterschiede in ihrer Resistenz gegenüber Parasiten zeigen. Diese genetischen Unterschiede stellen die basis für die evolutionäre Entwicklung durch die von Parasiten hervorgerufene selektion dar. Sowohl biotische als auch abiotische Umwelteinflüsse können die Expression bestimmter Eigenschaften von Wirten und Parasiten beeinflussen, und damit letztendlich auf die Fitness beider Gegenspieler einwirken. Einige Faktoren sind dabei wichtiger für die Fitness der Wirte, während wiederum andere die Fitness des Parasiten stärker einschränken.
